贺佳勤（秦海璐饰）与律师男友杨选（郭晓东饰）相恋五年后，感觉没有了初恋时的激情和冲动，自从邂逅摄影师孙祈伟（李亚鹏饰）后，她似乎找到了内心真正要找的感觉，而失恋的杨选则陷入迷惘之中。林菊若（马伊俐饰）看到每每喝醉的邻居杨选，都悉心照顾他，但杨选心中始终忘不掉佳勤。菊若被杨选的痴心感动，她想自己的男友也这么痴情该多好。贺佳勤的豪放不羁与孙祈伟的桀骜不驯像两块相互碰撞的磁铁，经过几番磨擦之后，终于粘在了一起。菊若不甘心看杨选消沉下去，可又不知道该怎样帮他。就在祈伟准备情定佳勤时，他的前女友不合时宜地出现了......。山田日後承認在開拍此片之前，已心諳渥美清的肺病步入末期，決定此章為最後一章，並安排由淺丘瑠璃子演的莉莉送寅次郎踏上人生的最後一程。分別15年後，兩人於如詩如畫的奄美大島重逢，感情依舊，鬥氣繼續，沒完沒了似的。正如山田所想，雖然系列必須完結，但始終不想為寅次郎的故事／人生劃上句號。結局如常開放，代表寅一直繼續他的旅程，日本人依然有夢。 公映後翌年，渥美清病逝，享年68歲，本片亦成為他的遺作。 Yamada knew before production began that Atsumi's health has deteriorated to such an extent that Tora-san to the Rescue would be the last film in the series.
